1. Place the purulent urethral dischar
sterile loop. Spread the specimen as thin as possible over the entire slide. If the specimen
is collected on a swab, roll the swab over a microscopic slide ( do not rub) so that all portions
of the swab head come in contact with the slide and the pus cells are distributed
in layers. This allows the characteristic intracellular organisms (within WBCs) to be
observed.
2. Gram stain by the routine procedure.
3. Examine the smear under microscope, under the oil-immersion objective. Pay partic
ular attention to edges of the smear, where cells are spread more thinly and are easier
to see, and the stain is less concentrated. Look for pus cells, and the presence of oval,
kidney-shaped, pale pink-coloured (Gram- negative) diplococci with adjacent sides at
tened (coee bean shaped), which are extracellular in the early stages, and intracellular
in acute conditions.
4. Report as follows:
