Observation
extent to which nuclei extend into the tail tip. Mansonella spp. does not have a sheath and the
tail is straight with a rounded tip. While examining, try to locate several microlariae so that
errors may not be incurred in case the sheath is torn or the tail is broken.

Bloodstaining procedure for examining microlaria
1. Prepare a thick smear of blood taken from a nger stick (no anticoagulant necessary) or
anticoagulated venous blood.
2. Air dry it.
3. Place the slide vertically in the staining trough (or beaker) lled with clean water. Wait
for 10 min for complete haemolysis.
4. Take slides out, drain them and air dry.
5. Stain for 30 min using a 1:10 dilution of Giemsa:
(a) Dilute Giemsa stain by mixing 9 mL of buffered water with 1 mL of stock stain solu
tion in a graduated cylinder. Prepare fresh.
