Preparation
1.Dissolve agar in 1000 mL of distilled water by heating.
2.While hot, add peptone and dextrose.
3.Boil gently until dissolved.
4.Adjust the pH to 6.0.
5.Dispense into culture tubes (20 mL) with cotton plugs or caps.
6.Sterilize by autoclaving (121 °C for 15 min).
