(PNS) are the cause of prominent neurological symptoms including
weakness, sensory loss, pain and autonomic dysfunction associated with
deficits, morbidity and mortality. These disorders may be primary
hereditary or cryptogenic neurologic disorders confined to the PNS or part
of the pathology of both the central nervous system and the PNS. Most
PNS disorders are secondary to other system disorders and may be
responsive to treatment of the primary disease. Important advances have
been obtained in several areas including molecular genetics, biochemistry,
immunology, morphology and physiology that have enhanced our
understanding of the causes and consequences of damage to peripheral
nerve. Understanding of both these groups of PNS diseases has greatly
expanded over recent years and has led to important advances of treatment
both to protect and to repair damages of peripheral nerve. This volume
provides an overview of the state-of-the-art of examination, diagnosis and
treatment of these very diverse disorders and will be of interest to both the
research and clinical neuroscience and neurology communities"--

disease (AD) in the quest for a cure, to date little progress has been made in
developing viable treatments for AD. Recent studies attempting toto determine
the initiating event of AD (β-amyloid plaques vs. neurofibrillary tangles) may
be interesting from a scientific perspective, but they do not seem to aid in
finding a cure. Perhaps more effort should be expended on effects of chronic
cerebrovascular diseases in elderly, such as hypertension and
hypercholesterolemia, which have been shown to result in premature cognitive
decline. Unfortunately, there are no studies that have determined the optimal
levels of blood pressure and lipids for cognition across the lifespan. Another
issue concerns the opinion expressed by some investigators that only
“validated” imaging techniques should be used for documenting
neurodegeneration in MCI and AD. Does this stifle progress toward finding
novel methodologies for documenting neurodegeneration in preclinical AD?
These issues are explored herein.

Amyloidoses are a heterogeneous group of diseases characterized by the
deposition of fibrillar protein in extracellular tissue space. Amyloid deposits
consist of a stable, insoluble aggregate of precursor proteins that have adopted
an antiparallel beta pleated sheet configuration, having an amorphous and
eosinophilic appearance. Although the reason for the accumulation of fibrils is
poorly understood, several factors may be involved seeing as there are certain
characteristics in precursor molecules that make them amyloidogenic, thus
predisposing them to amyloid fibril formation. Different types of amyloidosis
exist depending on the fibrillar protein precursor. The most common are: AL,
which is associated with plasma cell dyscrasia; AA, associated with chronic
