Discussion. Zoledronic acid is formed from the carboxylic acid, 2- (imidazole- 1- yl)acetic acid. (There are many options for the phosphorus reagent(s) and the solvent or diluent for this reaction. List the options and the yields associated with each option.) The carboxylic acid is formed by hydrolysis of the methyl ester. Methyl 2- (imidazole- 1- yl)acetate is formed by chloride displacement from methyl chloroacetate by imidazole.

Amiodarone Synthesis Ideas
An aromatic ketone is often formed by Friedel–Crafts Acylation. A phenyl ether is often formed by alkylation of a phenol. An ortho- iodophenol is often formed by iodination of a phenol. A phenol is often formed by demethylation of a methyl ether. A 2- substituted benzofuran is often acylated at C3.

Starting material salicylaldehyde is in Appendix A. Starting material methyl 2- bromohexanoate could be added to
Appendix A. Yields are high for the bromide displacement (99%) and ester hydrolysis (98%) [8]. The reaction of
2- (2- formylphenoxy)hexanoic acid with acetic anhydride likely affords 2- butylbenzofuran in 50–60% yield[9–11].
Methyl 2-
bromohexanoate is formed by bromination of hexanoic acid (Hell–Volhard–Zelinski Reaction) followed by
ester formation. Hexanoic acid is produced by fermentation.

Pros and Cons for Routes A and B to 2- Butylbenzofuran
Early in the course a pros and cons discussion may begin with the overall yield, number of steps, reaction times, and costs for starting materials. As the course progresses, other topics including physical properties of starting materials, reaction and workup solvents, throughput, safety, and environmental impact of byproduct(s) can be added.

Relative costs for carbon- containing starting materials:
A) phenol, chloroacetonitrile, 1- chlorobutane
salicylaldehyde, methyl 2- bromohexanoate
B)
Salicylaldehyde is made from and more expensive than phenol. One reference price for methyl 2$30/mol (Molbase, April 2020). This price, price estimates ($/mol) for chloroacetonitrile and chlorobutane, and a comparison of the routes to produce methyl 2- bromohexanoate, chloroacetonitrile, and 1- chlorobutane suggest that the starting material costs are lower for Route A than Route B. Based on lower costs for carbonthe preferred route.
