67.1 Introduction

Retinal ischemia/reperfusion (RI/R) injury is one of the common ocular diseases in clinical medicine, mainly seen in the ocular ischemic diseases caused by retinal vascular occlusion in such cases as central retinal vein or artery occlusion, acute angel-closure glaucoma and diabetic retinopathy. Its symptom is that many patients, after the reperfusion, find their retina seriously damaged and their visual function worsened even further. RI/R is the result of multiple factors, such as lack of energy, intracellular calcium overload, the exploding growth of free radicals, the excessive release of excitatory amino acid and the lack of neurotrophic factors, which turns on the gene monitoring apoptosis, leading to the apoptosis of retinal neural cells (Neal et al. 1994; Romano et al. 1993).

3 -daidzein sulfonate sodium (DSS) is a newly compounded and strongly watersoluble substance, produced by modifying the structure of daidzein, the main active ingredient of the Chinese herbal medicine Radix Puerariae. Earlier researches of our team have shown that DSS can help an organism anti-hypoxia, expand blood vessels and anti-oxidation (Zeng et al. 2005; Li, Zeng and Qiu 2005). Can DSS alleviate RI/R injury by enhancing an organism’s anti-oxidative ability? To find the answer, we produce RI/R models through the method of common carotid artery occlusion, so as to examine the protective effect of DSS on RI/R injuries, and explore the relationship between its protective effect and the anti-oxidative ability.

67.2 Materials and Methods

67.2.1 Animals and Reagents

Sprague Dawley (SD) rats (male, 250 280 g, age: 8 10 weeks) were obtained from Jiangxi traditional Chinese medical college and maintained in the Center of Laboratory Animal Resources at Gannan Medical College.

DSS was a white crystal powder produced by Shenyang Pharmaceutical University, with 99% purity. It is diluted to the needed concentration with NS before animal experiment. LDH, SOD and GSH-PX reagent kits were bought from Nanjing Jiancheng Biological Engineering Company.

67.2.2 Induction of RI/R

Twenty-four male SD rats, weighing 250–280 g, were randomly divided into four groups (n = 6/group), including sham control group, model group of RI/R injury, low dosage DSS (1 mg/kg) group and high-dosage DSS (2 mg/kg) group. The animals were anaesthetized with 10% Chloral hydrate solution (350 mg/kg, ip). Then, RI/R injury was induced by occluding common carotid artery on the left side. First,
