66.3 Results

66.3.1 Morphology and Quantity Changes of Müller Cells

At PND15, RCS rats Müller cells had formed long processes spanning the entire thickness of the retina. This Müller cell arrangement became disorganized by PND60 and by PND90 the Müller processes had became hypertrophic and extended processes into the subretinal space; note this was not found in control retinas. Müller cell counts at PND60, 90, 120, showed that the number of cells in RCS rats retinas were significantly higher than control (P < 0.05) (Fig. 66.1).

66.3.2 Expression of GFAP and ERK in RCS Rat Müller Cells

GFAP (glial fibrillary acidic protein) and ERK (extracellular signal – regulated kinase) are the activation-related proteins of Müller cell. At early age, the stain of GFAP and ERK in Müller cells was rare, with the progression of retinal degeneration, the stain of GFAP and ERK increased dramatically (Figs. 66.2 and 66.3). Western blots showed that at PND-30, 60, 120, the expression of GFAP and ERK in RCS rat retinas(GFAP:0.24±0.06, 0.32±0.07, 0.33±0.06; ERK: 0.11±0.03, 0.17±0.04, 0.18±0.04) was significantly higher compared with control(GFAP: 0.19±0.05, 0.24±0.04, 0.23±0.06; ERK: 0.08±0.03, 0.11±0.04, 0.11±0.03) (P < 0.05; Figs. 66.2–66.4).
