57 Effect of PBNA on the NO Content and NOS Activity in I/R Injury

treated PBNA group. Retinal I/R injury were induced by reperfusion followed occlusion of common carotid artery. Rats were anesthetized by peritoneal injection of 350 mg/kg 10% chloral hydrate. Sterilize the skin with 75% alcohol and made a midline incision in the ventral surface of the neck. Dissected muscles and lamina praetrachealis, to expose and isolate the comman carotid artery (CCA), and then occlude it with a clip. To make retinal ischemia sure, the pupils were dilated and fluorescence fundus angiography (FFA) were performed. After occlusion for 60 min, unclamp the clip and FFA were performed again to make sure the restoration of blood supply. Repeat all the steps for the sham operation group, except for occluding the common carotid artery. 1 ml/kg NS was injected through sublingual vein after CCA was dissociated in control group. Other groups were treated with normal saline, low-dosage PBNA (0.3 mg/kg) or high-dosage PBNA (1 mg/kg) through sublingual vein respectively before occlusion of CCA.

57.2.3Detection of MDA and NO Concentration, SOD and GSH-PX Activity

After 60 min reperfusion, collect blood by abdominal caval vein. The supernatants were collected by centrifugation at 3,000 r/min at 4C for 10 min and were stored at –80C. NO content was determined using Nitrate Reductase method, the activities of T-NOS, iNOS and eNOS were measured by chromatometry.

57.2.4 Statistical Analysis

All experimental data are expressed as means ± SEM. Statistical comparisons between the control and treatment groups were carried out using one-way analysis of variance (ANOVA) followed by Newman-Keuls post-hoc test. Probability levels of 0.05 or smaller were used for reporting statistical significance.

57.3 Results

57.3.1 Effect of PBNA on Serum NO Content in Retinal I/R Injury

Serum.NO content in I/R group showed a significant higher value than in the control group (P<0.001). Serum NO content rose in group treated with low-dosage PBNA (P<0.05) (Fig. 57.1).

57.3.2 Effect of PBNA on T-NOS Activity in Retinal I/R Injury

In both groups of low-dosage and high-dosage PBNA treated, T-NOS activities increased (P<0.01) (Fig. 57.2).

57.3.3 Effect of PBNA on iNOS Activity in Retinal I/R Injury

Serum iNOS activity in I/R group increased compared with control group (P<0.05), and declined after low-dosage PBNA treated (P<0.05) (Fig. 57.3).

57.3.4Effect of PBNA on Serum eNOS Activity in Retinal I/R Injury

Serum iNOS activity in I/R group was lower obviously than control group (P<0.05) and grew higher in groups treated with both low-dosage and high-dosage PBNA (P<0.05, P<0.01) (Fig. 57.4).
