57.1 Introduction

Retinal ischemia/reperfusion (I/R) injury is a clinical common ischemic optical disease, led to frequently by the occlusion of retinal vessels induced by the embolism of central retinal vein and artery, acute angle-closure glaucoma and retinopathy. In many patients, retina was seriously injured and visual function was further injured after the reperfusion followed occlusion. multiple factors such as Ca2+ overloading (Torin et al. 2000), free radical (Pannicke et al. 2005), the over release of exitotoxicitic amino acids (Kwon et al. 2005; Dijk et al. 2004), Inflammatory reaction (Sanchez et al. 2003), retinal nerve cell apoptosis (Oz et al. 2005), the changes of content of NO and activity of NOS (Cheon et al. 2003; Hangai et al. 1996), play roles in retinal I/R injury which can grow blind and can led to irreversible visual loss if treatment is not given promptly. So, it is very important to research on some effective new drugs to treat the retinal I/R injury.

Polygonum Bistora L, which is the dry rhizoma of Polygonaceae Plants, has the effects of clearing heat and eliminating dampness, relieving convulsion, detumescence and relieving pain. Extracts from Polygonum Bistora L were extracted by different solvents in turns: 95% alcohol, ligarine, acetic ether and n-butanol, and the n-butanol extracts were called Polygonum Bistorla L. n-Butyl Alcohol Extract (PBNA) which major reconstituents were ligustrin, quercetin-3-rutinoside, catechin and Mururin A (Liu et al. 2006). Our previous study showed that PBNA can expand blood vessels and protect heart muscle from ischemia (Li et al. 2007; Ye et al. 2006). To learn whether PBNA has a beneficial effect on retinal I/R injury, in this study we investigated the effect of PBNA on retina and the change of the content of NO and the activity of NOS and their relation in I/R injury by occlusion of common carotid artery.

57.2 Materials and Methods

57.2.1 Animals and Reagents

Sprague Dawley (SD) rats (male, 250 280 g, age: 8 10 weeks) were obtained from Jiangxi traditional Chinese medical college and maintained in the Center of Laboratory Animal Resources at Gannan medical college.

PBNA supplied by Shenyang Pharmaceutical University was dissolved in double distillation Water and 0.22 μm filtered. Kits of NO, T-NOS, iNOS and eNOS were provided by Nanjing Jiancheng Bioengineering Corporation.

57.2.2 Induction of Retinal I/R

Twenty-four rats were divided into four groups randomly: control group (sham operation), retinal I/R injury group, low-dosage PBNA treated group and high-dosage
