48.2.5 Statistical Analysis

All values represent the % mean of control. Differences between two groups of data were tested by a Student ‘t’ test. A p < 0.05 was used to assess significant differences between two groups.

48.3 Results

48.3.1 Effect of Glucose on 14C-CTGF Synthesis in hRPE Cells

Hyperglycemia (glucose = 20 mM) stimulated 14C-CTGF synthesis slightly in hRPE cells from control eyes obtained from Non-AMD patients and to a much greater extent in hRPE cells from a patient with AMD (Fig. 48.1).

48.3.2 Effect of IGF-1 on 14C-CTGF Synthesis in hRPE cells

IGF-1 (25 nM) stimulated 14C-CTGF synthesis minimally, if at all, in hRPE cells from eyes obtained of Non-AMD patients and to a much greater extent in hRPE cells from a patient with AMD (Fig. 48.2).

48.3.3Effect of PD98059 on Glucose Stimulated 14C-CTGF Synthesis in hRPE Cells

As shown in Fig. 48.3, elevated glucose (20 mM) stimulated 14C-CTGF synthesis only slightly in hRPE cells from Non-AMD control patients and to a much greater extent in hRPE cells from a patient with AMD, as seen in Fig. 48.1. However, PD98059 completely inhibited the glucose stimulated 14C-CTGF synthesis in Non-AMD patients. In addition, PD98059 (50 μM) only partially (but statistically significantly) inhibited glucose (20 mM) stimulation in hRPE cells from the AMD patient.

48.3.4Effect of PD98059 on IGF-1 Stimulated 14C-CTGF Synthesis in hRPE Cells

As shown in Fig. 48.4, the IGF-1 (25 nM) stimulated 14C-CTGF synthesis in hRPE from AMD as well as Non-AMD patients, shown in Fig. 48.2, is significantly inhibited by PD98059 (50 μM) in hRPE from Non-AMD patients. However, PD98059 (50 μM) did not inhibit IGF-1 (25 mM) stimulated CTGF synthesis in hRPE cells from the AMD patient.
